The mammalian matrix metalloproteinases (MMPs) degrade extracellular matrix in physiological and pathological processes. Of the sixteen members of matrix metalloproteinase family ten exist in soluble form whereas MT MMPs exist as integral membrane proteins. MMP15, first described as MTMMP2, is a metalloproteinase with a type I transmembrane domain. Initially reported in lung tissue, MMP15 message was also seen in testis, intestines, colon, heart, liver, kidney, pancreas and muscle. Western blots showed bands at 72 kD, 48 kD and 44 kD. Subsequent publications have focused on message, and found MMP15 in a wide range of tumor cell lines, as well as the synovial membrane from patients with rheumatoid arthritis. MMP15 is usually found in much lower levels than the more ubiquitous MMP14 (MT1MMP). MMP15 may be shed into the culture media when activated, but it is unclear if the shed form is proteolytically active. Cell lysates are the best source for Western blotting of MMP15.
