The elastin antibody I bought is ab77804 and I got a replacement that is ab21610. I tried the citrate buffer ph6 and antigen retrieval method that is recommended for this antibody:(In many cases, antigenicity of elastic fiber components can be enhanced by treating with 6 M guanidine HCl, 50 mM dithiothreitol, 20 mM Tris pH 8.0 for 15 minutes. After washing with 20 mM Tris pH 8.0, the slides are treated with 100 mM iodoacetamide in the dark for 15 minutes, washed and stained using normal protocols.)However, both of them didn't work out.

Thanks for letting me know. I am not able to get ab6310 to work out on my paraffin embedded sections. We just ordered ab7778 last week to see how it goes. And regarding to the elastin antibody (ab21607), the staining had alot of background (pretty much everywhere) and it didn't stain where the elastin should be at (I have skin and lung for control) even I followed the antigen-retrieval step as suggested on website.
I will let you know if whether ab7778 works this time, butI would actually like to see if we can get a refund on the elastin and collagen III antibody that we ordered.

I am sorry to hear that both of these antibodies have not given good results. I don't have any notes about the elastin antibody ab21607, have you used it with the same protocol as ab6310? Thank you for your phone call earlier this month and for letting us know about the trouble with these antibodies. We had discussed the IHC protocol that the lab used to test ab6310, and I have just received further information about this from the lab.

Ab6310 is only tested on unfixed frozen sections in-house, and the antigen retrieval method is 0.1% pronase for 10 minutes. However we also have feedback from other researchers using this antibody successfully in IHC on FFPE tissue, and heat-mediated antigen retrieval for 20 minutes in pH 6 citrate buffer should work as well. As I recall, you have already tried similar heat-mediated antigen retrieval, and I would be happy to send a replacement collagen III antibody if you would like to try a different one.
