Dear Tech support

One of our customers is interested in an anti PEX antibody, Pex is a MMP2 processing product present in different tumor types.
Searching for Pex gave 2 candidate antibodiesin Abcam's site - ab38922- against Hemopexin and ab79781 - against MMP2 C-terminal domain.

Both antibodies blot's show a band approximately 70Kda in size corresponding to the complete MMP2 protein but no a specific Pex band was observed
Should there be a Pex band on the Gel and what would be it's size? what would be the best antibody for Human Pex detection in WB?

The PEX domain of MMP 2 corresponds to amino acids 445-660 of the human protein sequence. The immunogen peptide use to create ab79781 corresponds to amino acids 643-660 of the human sequence. So ab79781 should be a useful antibody to specifically detect the PEX domain.

UniProt states "Autocatalytic cleavage in the C-terminal produces the anti-angiogenic peptide, PEX. This processing appears to be facilitated by binding integrinv/beta3." So perhaps PEX domain cleavage does not always occur and may only occur due to certain signaling events, and then only in certain cell types. I suggest checking the literature on PEX cleavage for further details.
