Western blot - MMP22 antibody (ab39082)
Our Abpromise guarantee covers the use of ab39082 in the following tested applications.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
WB: Recommended starting dilution of 1/1000 when using colorimetric substrates such as BCIP/NBT and 1/5000 for chemiluminescent substrates. Higher concentrations of antibody may be needed for non human samples. Detects bands of approximately 70kDa and 66kDa when used against the reduced protein. It is unclear if this represents alternative splicing, post translational modifications, aggregation, or some other phenomenon. (Predicted molecular weight: 53 kDa). Dilution optimised using Chromogenic detection. Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.
Protease. May regulate the surface expression of some potassum channels by retaining them in the endoplasmic reticulum.
Predominantly expressed in ovary, testis and prostate.
Belongs to the peptidase M10A family.
Contains 1 Ig-like C2-type (immunoglobulin-like) domain.
The toxin-like domain associates with, and blocks several potassium channels in the nanomolar to low micromolar range. The relative affinity is Kv1.6 > Kv1.3 > Kv1.1 = Kv3.2 > Kv1.4.
N-glycosylated.
Proteolytic cleavage might yield an active form.
Endoplasmic reticulum membrane. Membrane. A secreted form produced by proteolytic cleavage may also exist.

Western blot - MMP22 antibody (ab39082)

All lanes : Anti-MMP22 antibody - Propeptide domain (ab39082) at 1/1000 dilution
Lane 1 : Cell media from human fetal fibroblasts.
Lane 2 : Mouse lung tissue.
Predicted band size : 53 kDa
Observed band size : 66 kDa (why is the actual band size different from the predicted?)
Additional bands at : 70 kDa. We are unsure as to the identity of these extra bands.
It is unclear if the actual bands seen are from alternative splicing, posttranslational modifications, aggregation, or some other phenomenon.
ab39082 has not yet been referenced specifically in any publications.
