Application notes

Recommended dilutions WB: Recommended starting dilution of 1/1000 when using colorimetric substrates such as BCIP/NBT and 1/5000 for chemiluminescent substrates. Higher concentrations of antibody may be needed for non human samples. Detects bands of approximately 70kDa and 66kDa when used against the reduced protein. It is unclear if this represents alternative splicing, post translational modifications, aggregation, or some other phenomenon. (Predicted molecular weight: 53 kDa). Dilution optimised using Chromogenic detection. Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Relevance
|
MMP22 (or MMP23B) is a member of the matrix metalloproteinase (MMP) family, and it is part of a duplicated region of chromosome 1p36.3. Proteins of the matrix metalloproteinase (MMP) family are involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development, reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis.

All lanes : MMP22 antibody - Propeptide domain (ab39082) at 1/1000 dilution
Lane 1 : Cell media from human fetal fibroblasts. Lane 2 : Mouse lung tissue.
Predicted band size : 53 kDa Observed band size : 66 kDa (why is the actual band size different from the predicted?) Additional bands at : 70 kDa. We are unsure as to the identity of these extra bands.
It is unclear if the actual bands seen are from alternative splicing, posttranslational modifications, aggregation, or some other phenomenon.
