Ab11647 specifically binds to MMP24 and does not cross-react with the other MMP family members (MMP1, MMP2, MMP3, MMP9). The cytoplasmic domain of MMP24 is detached when MTMMPs shed from the membrane, thus this antibody should not detect these forms of MMP24.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
WB: A recommended starting concentration for Western blots is 1:2,000 when using colorimetric substrates such as BCIP/NBT and 1:5000 for chemiluminescent substrates. Higher concentration of antibody may be needed for non-human samples The antibody identifies a band at 65 kD (zymogen) and also the activation and breakdown products. Ab11647 also recognizes non-reduced (native) MMP24. Note: Since cell types differ greatly in the quantity of MMP24 produced, the conditioned media may require mitogen stimulation to visualize the bands by Western blotting. Treatment of cells with concanavolin-A or the phorbol ester TPA stimulates production of MMP24 in some cell types, and the enzyme can be recovered in cell lysates. Dilution optimised using Chromogenic detection. Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.
Matrix Metalloproteinase 24 (MMP24, MT5MMP) has been described as a proteoglycanase, which is critical in the turnover of ECM (extracellular matrix) components in the brain. MMP24 contains a candidate signal sequence, a propeptide region, a potential furin recognition motif, a zinc-binding catalytic domain, a hemopexin-like domain, a 24-residue hydrophobic region, which acts as a potential transmembrane domain, and a short cytosolic domain. Unlike other MTMMPs, MMP24 (MT5MMP) sheds from the cell surface as soluble proteinases, allowing its versatility as both a cell bound and soluble proteinase in ECM remodeling processes. MMP24 is elevated in several tumor cell lines, and is also produced by some normal cell lines. It is predominantly expressed in the brain, indicating a role in nervous system development. MMP24 is also highly expressed during embryonic development. The mouse MMP24 gene maps to chromosome 2.
