Specificity

ab38914 recognises the Aminoterminal end of active enzyme MMP3 (stromelysin 1) and Stromelysin 2, but does not cross react with Stromelysin 3, nor to the other MMP family members (MMP 1, MMP 2, MMP 9, etc.).

Application notes

WB: 1/1000, when using colorimetric substrates such as BCIP/NBT - 1/5000, when using chemiluminescent substrates. Predicted molecular weight: 54 kDa. But note that when used against the reduced protein, ab38914 identifies bands at 59 Kd and 57 Kd (the pro-form), as well as active forms. Also note that Stromelysin levels in quiescent cells and tissues are minimal, and stimulation or protein concentration is often needed to visualize the bands. Dilution optimised using Chromogenic detection. Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Can degrade fibronectin, laminin, gelatins of type I, III, IV, and V; collagens III, IV, X, and IX, and cartilage proteoglycans. Activates procollagenase.

Sequence similarities

Belongs to the peptidase M10A family.
Contains 4 hemopexin-like domains.

Domain

The conserved cysteine present in the cysteine-switch motif binds the catalytic zinc ion, thus inhibiting the enzyme. The dissociation of the cysteine from the zinc ion upon the activation-peptide release activates the enzyme.

Secreted > extracellular space > extracellular matrix.
