Specificity
ab38913 recognises the propeptide of MMP3 (Stromelysin 1) and Stromelysin 2, but does not cross react with Stromelysin 3, nor to the other MMP family members (MMP1, MMP2, MMP9, etc.).
We have a range of domain specific antibodies for this target. For a full list please see all MMP3 antibodies

Application notes

Recommended dilutions WB: 1/1000, when using colorimetric substrates such as BCIP/NBT - 1/5000, when using chemiluminescent substrates. Predicted molecular weight: 54 kDa. But note that when used against the reduced protein, ab38913 identifies bands at 59 Kd and 57 Kd (the pro-form), and not the active forms. Also note that MMP3 levels in quiescent cells and tissues are minimal, and stimulation or protein concentration is often needed to visualize the bands. Dilution optimised using Chromogenic detection. Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Relevance
Proteins of the matrix metalloproteinase (MMP) family are involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development, reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis. Most MMP's are secreted as inactive proproteins which are activated when cleaved by extracellular proteinases. MMP3 is an enzyme which degrades fibronectin, laminin, collagens III, IV, IX, and X, and cartilage proteoglycans. The enzyme is thought to be involved in wound repair, progression of atherosclerosis, and tumor initiation. MMP3 is reported to be implicated in rheumatoid arthritis and some forms of cancer.
