I am looking for an antibody specific for the active form of human MMP-8. ab38992 says it is specific for the amino terminal end of active MMP-8 which would lead one to believe that the antibody does not react with pro-MMP-8. However, ab11609, also raised against the N-terminal peptide, reacts with active forms of MMP-8 (64kDa and 58kDa) as well as the non-reduced MMP-8 (i.e. the pro-MMP-8). So, is it correct to think that neither of those antibodies is really SPECIFIC for active MMP-8 (by specific I mean that the antibody does not react with the pro-MMP-8)?

Thank you for your enquiry and your interest in our products. Both antibodies recognize the active form(s) of MMP-8, however the imunogen sequences used to develop these two antibodies were different. On the datasheet of ab11609 the note saying - it reacts with active forms as well as the non-reduced MMP-8 - which implies that ab11609 does work under reducing (in the present of SDS, DTT, beta-mercaptoethanol) and under non-reducing conditions. This does not mean that ab11609 does recognize the pro-form. I hope this will be useful for you. |
