Hello, I've tried to get the MMP-9 sELISA to work using the antibodies and amounts you gave below but I've found that there's interactions between the two antibodies without the protein, have you heard of this before and if so, is there anything I can do to reduce this? Thanks. |

ANSWER: |
Thank you for contacting us and for alerting us to the problem you are experiencing with our products. I am sorry to hear you have had difficulty obtaining satisfactory results from these MMP9 antibodies. We take product complaints very seriously, and investigate every product that we feel may not be performing correctly. However, we have not heard about this problem with the MMP9 antibodies before. I have attached the protocol which has been used to test the antibodies and I hope this is of help to you. If the issue persists and/or your protocol is not significantly different from the one attached, I would like to investigate this particular case further for you. In order to do this and also obtain some further information for our quality records (e.g. lot numbers, conjugate of secondary antibodies, substrate etc.), I have enclosed a questionnaire and would appreciate if you could complete this. It will help you put the information we require together very easily. I would appreciate if you could provide some data that illustrate the problem (e.g. readings for the various controls). We are happy to offer you tailored technical support, and if it turns out that the products are not functioning in the applications cited on the product data sheet, we will be pleased to provide a credit note, free of charge replacement or refund. I hope this information is helpful, thank you for your cooperation. Should the suggestions not improve the results, please do not hesitate to contact me again with the further requested details. |

I am looking for an antibody against MMP9 and TIMP1 for WB in pig. As there is no tested antibody as yet, I would be grateful if you could send me the immunogen sequence of this two antibodies to allow me to do a homology search. Thanks. |
|||||||||
ANSWER: |
Thank you for your enquiry. The immunogen sequence used to raise ab35399 was LQGSEKGFQSRHLA; corresponding to amino acids 175-198 of Human TIMP1. The immunogen sequence used to raise ab38902 is confidential. However, if you provide me with the accession number of the pig homolog I can certainly request that the lab performs a sequence comparison allowing you to predict the likely reactivity of this antibody. I hope this information helps, please do not hesitate to contact us if you need any more advice or information. |

Please note: All products are "FOR RESEARCH USE ONLY AND ARE NOT INTENDED FOR DIAGNOSTIC OR THERAPEUTIC USE"
Standard curve for MMP9 (Analyte: ab82955); dilution range 1pg/ml to 1µg/ml using Capture Antibody Mouse monoclonal [SB15c] to MMP9 (ab51203) at 5µg/ml and Detector Antibody Rabbit polyclonal to MMP9 - Hinge region (ab38902) at 0.5µg/ml.
All lanes : Anti-MMP9 antibody - Hinge region (ab38902)
Lane 1 : Human MMP9
Lane 2 : Cell media from human chondrosarcoma (untreated)
Predicted band size : 78 kDa
Observed band size : 92 kDa (why is the actual band size different from the predicted?)
When used against the reduced protein, ab38902 identifies bands at 92kDa and 88kDa (the pro form and active form respectively).
