Do you think it's possible to use this antibody for FACS?
|
|||||||||
ANSWER: |
Thank you for getting back to me. Unfortunately I cannot really comment on whether this antiserum will work by FACs as it is yet to be tested. It is difficult to predict whether an antiserum will work by a given application. From the listed applications on the datasheet it seems a highly versatile antibody, and there is no reason to suggest that it shouldnt be tried by FACs. I would certainly be very interested in how it performs. If you decide to go ahead and purchase this product, please let us know how you get on by submitting an Abreview and in return we will award you 50 Abcam Points, which can be redeemed on a number of rewards (a further 100 Abcam Points will be offered for an image). |

I notice that the data sheet for this antibody states that it has been tested in flow cytometry and i was wondering if you could cite a paper or data that can confirm this. Many thanks |

ANSWER: |
Thank you for your enquiry. Unfortunately there was an error on the datasheet and according to my records this antiserum is yet to be tested by FACs. I have revised the datasheet to reflect this. Please may I apologise for the confusion. |

I am interested in your antibodies for BCRP (ab3380: Clone BXP-21), MDR1 (ab10333: Clone 4E3), MRP2 (ab3373: Clone M2 III-6) and MRP3 (ab3375: Clone M3II-9). As I will be using these antibodies for FACS, I would like to know whether you have any of these in a conjugated version (FITC or PE) as well? If not, do you have any matching secondairy antibodies which are conjugated with FITC or PE? Are these antibodies available at the moment? If I buy one vial of each of these antibodies in one go, will there be any special price offer? I would prefer to detect only the proteins which are present in the plasma membrane. As the MRP antibodies are reacting with an internal epitope, it will be necessary to permeabilize the plasma membrane. Do you know of any more or less selective technique to permeabilize the plasma membrane resulting in a selective staining of MRP2 and MRP3 in the plasma membrane only? I am lokoing forward to your reply. Thanks for your help!

ANSWER: |
Thank you for your enquiry and for your patience. Unfortunately, none of these antibodies are available in a conjugated form but we do have a wide range of secondary antibodies to choose from (including ones conjugated to FITC and PE). At the bottom of the online datasheets for each of these primary antibodies, there is a list of compatible secondary antibodies that can be used. We unfortunately can't offer any discounts. All the information that we have regarding these antibodies is located on the online datasheets, and ab10333 and ab3373 have been tested for application in FACS but ab3380 and ab3375 have not to our knowledge. If you have any more questions, please contact us again.

ab3373 at 1/200 staining rat liver tissue sections by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections). The tissue was formaldehyde fixed and then heat mediated antigen retrieval was performed using citrate buffer. The antibody was incubated with the tissue for 1 hour. An HRP conjugated goat anti-mouse antibody was used as the secondary.
