Due to our experience with previous antibodies we are concerned about the inability to measure the correct bands in the presence of unspecific binding.Do you have any positive controls for these two antibodies? If so, could we get a small sample of those positive controls if we purchase these two.Or, if we are unable to detect the correct bands for these two A.b. due to a lot of unspecific binding, is that backed up by the Abpromise guarantee? Thank you for your reply. I have checked our catalog and we do have a positive control available, human skeletal muscle whole cell lysate ab29330. We do not have samples available, but if the antibodies produced many non-specific bands in WB, this is covered by our Abpromise and you would be eligible for a replacement or refund per our guarantee. I am currently working on quantifying protein levels of the two ubiqutin ligases FBOX32 a.k.a MAFbx and MuRF-1 in human skeletal muscle using the Western Blot technique. Accordingly, I’m contacting you to see if you have any antibodies against these proteins that you know for sure have a good specificity. We are using homogenates from freeze-dried human skeletal muscle. The antibodies I have been looking at are your “Anti-Fbx32 antibody (ab67866)” and “Anti-MURF1 antibody (ab77577)”. Would these be recommended for WB of human skeletal muscle homogenates or do you have other alternatives? Do these antibodies have a good specificity? Since we have been having major trouble with all our previous Fbx32 and MuRF-1 antibodies we want to be confident of its specificity before we make our purchase. Is there any possibility for you to provide with a test sample of these antibodies so that we can try them out before we make or purchase? Do you have any positive controls for these proteins so that we can be sure that we are measuring the right bands?

Thank you for contacting Abcam regarding out FBX32 and MURF1 antibodies. I would like to recommend two products that will be suitable for your experiments. Regarding Anti-Fbx32, I would like to recommend ab92281. As shown on the online datasheet, a very specific single band is detected in human skeletal muscle lysate. Regarding an Anti-MURF1 antibody, I would like to recommend ab57865, as ab77577 is not in stock and may not be available for several months. This antibody was originally tested with the recombinant fragment immunogen peptide, and has since been shown to recognize endogenous protein in IHC. Please see our online datasheet for more information. IHC image of ab77577 staining in Mouse Skeletal Muscle formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab77577, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.
