Applications

Show applications key

Our Abpromise guarantee covers the use of ab77577 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Target

Function

E3 ubiquitin ligase. Regulates proteasomal degradation of cardiac troponin I/TNNI3 and probably of other sarcomeric-associated proteins. May play a role in striated muscle atrophy and hypertrophy by regulating an anti-hypertrophic PKC-mediated signaling pathway. May regulate the organization of myofibrils through TTN in muscle cells.

Tissue specificity

Muscle specific. Selectively expressed in heart and skeletal muscle. Also expressed in the iris.

Pathway

Protein modification; protein ubiquitination.

Sequence similarities

Contains 1 B box-type zinc finger.
Contains 1 COS domain.
Contains 1 RING-type zinc finger.

Developmental stage

Expressed throughout all developmental stages.

Domain

The RING-type zinc finger mediates interaction with SUMO2 and localization to the nucleus. Also required for the E3 ubiquitin ligase activity.

Cellular localization

Cytoplasm. Nucleus. Localizes to the M- and Z-lines in skeletal muscle. Colocalizes with TNNI3 in myocytes.

  Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - MURF1 antibody (ab77577)

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - MURF1 antibody (ab77577)

IHC image of ab77577 staining in Mouse Skeletal Muscle formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab77577, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.
