We have ordered a couple of weeks ago 3 Abs for matrin3, TRBP2 and Mov10 proteins. We used those Abs for WB and none of them worked. TRBP2 (AB72110) it doesnt give any band after o/n or 2-3h RT even in 1/500 dilution and matrin3 (ab51081) Mov10 (ab80613) they really give many many bands but not the right ones.

These Abs came together, is it possible that sth happen during transfer? Would you please help us with make them work? Alternatively, could you please consider change them with another lot that you know is working or another Ab for these mouse proteins?

But, it would have been nice if you send to me the other antibody now because this antibody is indeed for WB, though it is only predicted for mouse. I would have liked to do the experiments in the same day with the same sample.

I can understand why you would like to test both antibodies at the same time, and therefore I am willing to offer you this as a goodwill gesture. In order to do this I will need the original order number for ab70336. If you do not have this to hand, a delivery address and an approximate date of delivery should be enough. If you can get this information to me before 4 pm (Netherlands time) I will be able to set up the shipment to be with you tomorrow.

I would appreciate it though, if you do have trouble with the western blotting (particularly ab70336) if you could let me know prior to publishing the Abreview as there may be suggestions I can make in optimising the protocol.

I promised you to send an e-mail to you over the above mentioned product. As discussed, I thought that this antibody can be used for WB. It appears not. First, can you please tell me why it is not suitable for WB? Second, what solution can you provide me? The only other anti-matrin 3 of Abcam that may be used for WB is the product with the catalog No. Ab51081, even here there seems uncertainy because I want to use the anti-body against mouse. Even for the latter antibody, you indicate that it can be used for human, and that it is predicted for mouse and some other species. Nevertheless, at least it is predicted for mouse. The referenc that I told you yesterday: "Kula et al. Characterization of the HIV-1 RNA associated proteome identifies matrin 3 as a nuclear cofactor of Rev function" published in Retrovirology (2011) 8:60. On close examination, I noticed these authors used Ab70336 for immunoprecipitation and for the WB it seems they have used another antibody from VIVA.

Thank you for contacting us and I'm sorry in the delay in getting back to you.

As discussed over the phone yesterday, I am seeking more information from the source of this antibody to ascertain exactly why ab70336 was not deemed suitable for use in Western blotting. Once I have more information in this regard I will let you know.

Ab70336 has been used to detect both the mouse and human protein. This is the only anti-Matrin 3 antibody from our catalogue which has been used to detect the mouse protein. http://www.abcam.com/Matrin-3-antibody-ab51081.html, which has been validated in western blotting is however predicted to react with the mouse protein due to the sequence homology between the immunogen used to raise the antibody and the mouseMatrin 3 protein. These sequences share 98% homology (please refer to the attachment of this email). As mouse samples have not been tested with this antibody, I cannot however guarantee that it will.

I think I would suggest it would be worth trying ab70336 with your mouse samples in western blotting. If you find that it does not work then I can offer you ab51081 free of charge to try, if you will submit an Abreview for the two antibodies.This will only take 5 minutes to submit and would be invaluable data to our other customers looking for similar Matrin 3 antibodies as yourself.
