I have noted that the WB using ab49783 and ab42463 look quite different. Whiule it is indicated that the cell ty ped probed woth ab59783 is SK-HEP-1, the source of protein for the WB with ab 43463 is not given. Could you precise and comment about the differences between the 2 Ab? |

Thank you for your enquiry and your interest in our products. The sample used in the ab42463 Western blot image (correctly noted on the Abcam website) is a dilution of human milk. The epitope for ab42463 is towards the carboxyterminal end of the catalytic domain, and the epitope for ab59783 is closer to the aminoterminal end of the catalytic domain. I would like to know the difference between ab42463 and ab59783. I need a good antibody against matriptase 2 and matriptase-1. I want to look into lysat the presence of these proteins. Which antidobies do you recommend? |

Thank you for your enquiry. The difference between ab42463 and ab59783 is that the immunogen sequence is difference. Both immunogens are within the catalytic domain (amino acids 568- 802 of the 802 amino acid human Matriptase-2 sequence). The ab59783 epitope falls within the range of amino acids 610 - 690, and the ab42463 epitope falls within the range of amino acids 760 - 802; we cannot give out the exact amino acid sequence of any of our immunogenic peptides, because that information is proprietary. Both antibodies work well in western blotting. It seems that we currently do not have antibodies to matriptase-1 I'm afraid. Anti-Matriptase 2 antibody - Catalytic domain (ab42463) at 1/1000 dilution + Human Milk at 1/20 dilution
ICC/IF image of ab42463 stained HepG2 cells. The cells were 100% methanol fixed (5 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab42463, 1µg/ml) overnight at +4ºC. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
