Recommended dilutions ICC: 1/500. ICC/IF: 1/500 (see Abreview) IP: Use at a concentration of 2 - 4 µg/ml. WB: Use at a concentration of 2 µg/ml. This concentration detects membrin in 20µg of HeLa heat shocked cell lysate by ECL analysis. Predicted molecular weight: 27 kDa.
Not tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

|
Membrin is a 27 kDa integral membrane protein on the surface of the Golgi apparatus that serves as a tSNARE in ER to Golgi transport. GS27 is a member of a protein complex that consists of cis-Golgi vesicle receptor syntaxin 5 and GS28, vSNARE rbet1 and rsec22, and rsly1. Immunoprecipitation analysis has shown that there are two subcomplexes within this complex: one contains syntaxin 5 and GOS28 and the other one contains syntaxin 5, GS27/membrin, rsec22 and rbet1. These complexes are postulated to direct the fusion of ER-derived vesicles with Vesicular tubular cluster (VTC), and the fusion of VTCs to form cis-Golgi compartment.

Immunocytochemistry/ Immunofluorescence - Membrin antibody [4HAD6 ] (ab13511)
ab13511 at 1/500 dilution staining human HeLa cells by ICC/IF. HeLa cells, grown on coverslips, were fixed in 3% paraformaldehyde in PBS for 15 minutes. After washing with PBS, the reaction was quenched with 50 mM ammonium chloride for 10 mins. The cells were permeabilized using 0.1 % Triton X100 in PBS, for 2 minutes. After washing 2 times with PBS, the sample was blocked with 2% fish gelatine in PBS. for 1 hour. ab13511 was diluted in the blocking solution, put on the coverslips and incubated for 1 hour on room temperature. After washing 3-4 times with the blocking solution, the sample was incubated in the secondary antibody (Alexa Fluor ® 488 conjugated Goat Anti-Mouse IgG (H+L) diluted in the blocking solution), for 1 hour at room temeperature.
