ab13511 at 1/500 dilution staining human HeLa cells by ICC/IF. HeLa cells, grown on coverslips, were fixed in 3% paraformaldehyde in PBS for 15 minutes. After washing with PBS, the reaction was quenched with 50 mM ammonium chloride for 10 mins. The cells were permeabilized using 0.1 % Triton X100 in PBS, for 2 minutes. After washing 2 times with PBS, the sample was blocked with 2% fish gelatine in PBS. for 1 hour. ab13511 was diluted in the blocking solution, put on the coverslips and incubated for 1 hour on room temperature. After washing 3-4 times with the blocking solution, the sample was incubated in the secondary antibody (Alexa Fluor ® 488 conjugated Goat Anti-Mouse IgG (H+L) diluted in the blocking solution), for 1 hour at room temeperature. The coverslips were washed 6 times with the blocking solution and then mounted on microscope slides.
