Recommended dilutions IHC-P: 1/250. Perform heat mediated antigen retrieval before commencing with IHC staining protocol (See Abreview). WB: Use at a concentration of 1 µg/ml. Detects a band of approximately 52 kDa (predicted molecular weight: 52 kDa). Can be blocked with MiTF peptide (ab21454).
Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Relevance
|
MiTF (Microphthalmia associated transcription factor) is a basic helix loop helix leucine zipper (b HLH ZIP) transcription factor implicated in pigmentation, mast cells and bone development. Mutations in MiTF cause auditory pigmentary syndromes, such as Waardenburg syndrome type II, type IIa and Tietz syndrome in humans. There are two known isoforms of MiTF differing by 66 amino acids at the NH2 terminus. Shorter forms are expressed in melanocytes and run as two bands at 52kDa and 56kDa, while the longer Mi form runs as a cluster of bands at 60-70kDa in osteoclasts and in B16 melonoma cells (but not other melanoma cell lines), as well as mast cells and heart. MiTF plays a critical role in the differentiation of various cell types as neural crest-derived melanocytes, mast cells, osteoclasts and optic cup-derived retinal pigment epithelium.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - MiTF antibody (ab20663)
ab20663 staining MiTF in mouse skin tissue section by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections). Tissue section was fixed in formaldehyde and after heat mediated antigen retrieval, blocked (5 minutes/peroxidase block then 10 minutes/protein block) for 15 minutes at 20°C. The primary antibody was diluted, 1/250 and incubated with sample for 45 minutes at 20°C. A HRP conjugated goat polyclonal to rabbit IgG was used undiluted, as the secondary. ab20663 staining MiTF in meanoma tissue sections by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections). Tissue underwent formaldehyde fixation before heat mediated antigen retrieval and then blocked (5 minutes of peroxidase block and 10 minutes of protein block) for 15 minutes at 20°C. The primary antibody was used undiluted and incubated with sample for 45 minutes at 20°C. A HRP conjugated goat polyclonal to rabbit IgG was used, undiluted as secondary.
