Our Abpromise guarantee covers the use of ab36076 in the following tested applications.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
WB: Use a concentration of 1 µg/mlDetects a band of approximately 35 kDa (predicted molecular weight: 33 kDa).
ICC/IF: Use a concentration of 1 µg/ml
IHC-P: Use at an assay dependent dilution.
Adapter protein involved in the Toll-like receptor and IL-1 receptor signaling pathway in the innate immune response. Acts via IRAK1, IRAK2, IRF7 and TRAF6, leading to NF-kappa-B activation, cytokine secretion and the inflammatory response. Increases IL-8 transcription. Involved in IL-18-mediated signaling pathway.
Ubiquitous.
Defects in MYD88 are the cause of MYD88 deficiency (MYD88D) [MIM:612260]; also known as recurrent pyogenic bacterial infections due to MYD88 deficiency. Patients suffer from autosomal recessive, life-threatening, often recurrent pyogenic bacterial infections, including invasive pneumococcal disease, and die between 1 and 11 months of age. Surviving patients are otherwise healthy, with normal resistance to other microbes, and their clinical status improved with age.
Contains 1 death domain.
Contains 1 TIR domain.
The intermediate domain (ID) is required for the phosphorylation and activation of IRAK.

Western blot - MyD88 antibody (ab36076)

Anti-MyD88 antibody (ab36076) at 1 µg/ml + Jurkat (Human T cell lymphoblast-like cell line) Whole Cell Lysate at 10 µg
Secondary
IRDye 680 Conjugated Goat Anti-Rabbit IgG (H+L) at 1/10000 dilution
Performed under reducing conditions.
Predicted band size : 33 kDa
Observed band size : 35 kDa (why is the actual band size different from the predicted?)
Additional bands at : 41 kDa. We are unsure as to the identity of these extra bands.
Immunocytochemistry/ Immunofluorescence - MyD88 antibody (ab36076)

ICC/IF image of ab36076 stained MCF7 cells. The cells were 4% PFA fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab36076, 1µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
Western blot - Anti-MyD88 antibody (ab36076)

Anti-MyD88 antibody (ab36076) at 1 µg/ml + Lung (Mouse) Tissue Lysate at 10 µg
Secondary
Goat polyclonal Secondary Antibody to Rabbit IgG - H&L (HRP), pre-adsorbed (ab97080) at 1/5000 dilution
developed using the ECL technique
Performed under reducing conditions.
Predicted band size : 33 kDa
Observed band size : 33 kDa
Additional bands at : 65 kDa,70 kDa,85 kDa. We are unsure as to the identity of these extra bands.
Exposure time : 20 minutes
