Specificity
|
This antibody is specific to an epitope of amino acids 3-56 in the N-terminus of the 45 kDa mouse MyoD1 protein. This antibody does not react with myogenin, Myf5 or Myf6. MyoD1 stains the nuclei of myoblasts in developing muscle tissues. MyoD1 is not detected in normal adult tissue but is expressed strongly in the tumor cell nuclei of rhabdomyosarcomas.

Recommended dilutions IHC: This antibody may be diluted to a titer of 1:20-1:50 in an ABC method. Staining Protocol: We suggest an incubation period of 30-60 minutes at room temperature. However, depending upon the fixation conditions and the staining system employed, optimal incubation conditions and antibody dilutions should be determined by the user. Formalin fixed paraffin embedded tissue sections require heat mediated antigen retrieval with 10 mM citrate buffer, pH 6.0 prior to immunostaining.

Relevance
|
MyoD1 belongs to the basic helix-loop-helix family of transcription factors and the myogenic factors subfamily. It regulates muscle cell differentiation by inducing cell cycle arrest, a prerequisite for myogenic initiation. Myod1 is essential for repair of damaged tissue. It activates its own transcription which may stabilize commitment to myogenesis.
