Mouse monoclonal [GC-4] to N Cadherin

Specificity

Monoclonal Anti-N Cadherin/ACAM is specific for a polypeptide (135 kDa), isolated from a freshly prepared extract of rat or chicken cardiac muscle, when assayed by immunoblotting. This antibody reacts with the ACAM molecule in tissues and cells from human, chicken, rat, mouse, and rabbit.

Monoclonal Anti-N Cadherin/ACAM may be used for the localization of a 135 kDa receptor of the intercellular adherens junction in immunofluorescent microscopy, electron microscopy, and immunoblotting. This antibody can also be used to study the mechanism of adhesion and interaction of cultured cells and cells in tissues. Monoclonal Anti-NCadherin/ACAM stains a large variety of intercellular junctions formed by chicken fibroblastic and epithelial cells in culture. In adult mammals, the antibody stains predominantly heart and lens tissue and also stains a large variety of embryonic tissue. By double labelling, this antibody appears to localize vinculin and actin-rich intercellular contacts and is absent from cellular matrix attachment sites.

Cadherins are calcium dependent cell adhesion proteins. They preferentially interact with themselves in a homophilic manner in connecting cells; cadherins may thus contribute to the sorting of heterogeneous cell types. CDH2 may be involved in neuronal recognition mechanism. In hippocampal neurons, may regulate dendritic spine density.

ab11340, at 1/100, staining N cadherin in mouse embryonic stem cells by immunocytochemistry/ immunofluorescence. The cells were methanol/acetone (1/1) fixed and blocked with 1% serum and 0.1% BSA prior to incubation with the antibody for 30 minutes. Alexa fluor® 488 chicken polyclonal to mouse IgG, diluted 1/200, was used as the secondary antibody.
