Product type

Proteins and Peptides

Peptide

Recombinant full length Human NAT2 (derived from the wild type NAT2*4 allele) and expressed in baculovirus infected BTI insect cells.

Specificity

Unit Definition: One unit will acetylate one pmole of sulfamethazine per minute at pH 7.5 at 37 °C.

FuncS

Application notes
(see key)

Recommended dilutions
N-acetyl transferase 2 Activity:
Incubations were conducted at 0.5 µg/ml protein in 50 mM triethanolamine (pH 7.5), 1 mM EDTA, 1 mM DTT, 4.6 mM acetyl-d,l-carnitine, 0.06 U/ml carnitine acetyltransferase, 0.1 mM acetyl coenzyme A, and 40 µM sulfamethazine as substrate for 15 minutes at 37 °C. The reaction is stopped by the addition of 0.025 ml of 6 % acetic acid/94% acetonitrile.

Not yet tested in other applications.
Optimal dilutions/concentrations should be determined by the end user.

Relevance

NAT2 participates in the detoxification of a plethora of hydrazine and arylamine drugs. It catalyzes the N- or O-acetylation of various arylamine and heterocyclic amine substrates and is able to bioactivate several known carcinogens.

NAT2 is derived from a soluble cell fraction from insect cells (Trichopulsi ni, BTI-TN-5B1-4) expressing human NAT2 wild-type cDNA using a baculovirus expression system.
