Biological activity

Activity: 70 U/mg. One unit is defined as the amount of enzyme that will phosphorylate 1 nmol NEK6 substrate (FLAKSFGSPNRAYKK) per minute at pH 7.4 and 30C. Assay buffer: 50 mM HEPES, pH 7.4, 3 mM MgCl2, 3 mM MnCl2, 1 mM DTT, 3 uM Naorthovanadate, 0.1 mM ATP, 4 uM NEK6 substrate, and 0.4 mg/ml NEK6.

Activated during M phase. Required for chromosome segregation at metaphase-anaphase transition and therefore for mitotic progression. Inhibition of activity results in apoptosis. Phosphorylates KIF11 to promote mitotic spindle formation.
