Dear Abcam orders, Im looking to order antibody number ab83508. Altoght there is no reference published it has worked to stain oligodendrocyte progenitors in mouse brain, only a picture I could find. What does Tissue specificity section mean? 'Detected only in malignant melanoma cells.' We are staining mouse brain tissue and have some positive staining in localized regions, the staining looks like membrane stain. |

Thank you for your telephone enquiry yesterday afternoon. I can understand you concerns regarding the specificity statement on the datasheet. I would liketo confirm that this is a statement from the tissue specificity section in Uniprot. We have an agreement with Uniprot to link to the information from their pages for some sections of our datasheets. As you discussed with me, this is a rather confusing sentence. Because it mentions 'detected' it perhaps gives the impression that this particular antibody detects NG2 only in malignant melanoma cells. However, I can confirm that NG2 is expressed in a range of other tissues, according to the EST expression profile from Unigene. Including brain tissue: http://www.ncbi.nlm.nih.gov/UniGene/ESTProfileViewer.cgi?uglist=Hs.513044 I have also found the following publications online describing NG2 expression in mouse brain: http://dev.biologists.org/content/135/1/145.full http://www.ncbi.nlm.nih.gov/pubmed/20346197 I can suggest to try a further literature search for more information. I can confirm the concentration of ab66219 (Batch: GR9812) of this ab is 1 mg/ml. The other antibody ab36204 (Lot: GR42382) is not affinity purified but whole antiserum so the IgG is not quantified. However, I can provide a range of the IgG: 10-15 mg/ml.

Dear Sir or Madam, We recently bought the following antibodies from your company: ab36204; ab48614; ab66219; ab83508; ab102998. As we would like to test at least four of them on 4%PFA, sucrose fixed brain tissue and include the correct controls I have a few questions: I received the following list: ab36204 (Lot: GR42382) --> concentration: N/A 100µl ab48614 (Lot: GR62244) --> concentration: 0,7699 mg/ml (100µg) ab66219 (Lot: GR9812) --> concentration: N/A 100µl ab83508 (Lot: GR38090) --> concentration: 0.5 mg/ml (100µg) ab102998 (Lot: GR80395) --> concentration 0.25mg/ml (100µg) In order to include a control with the same antibody concentration as for the primary antibody it would be good to know the concentration of ab36204 and ab66219, too. Could you help me concerning that matter? Second question, do you have any experience with good control antibodies? I thought the following: ab36204 --> Rabbit IgG ab48614 --> Rabbit IgG ab66219 --> Rabbit IgG ab83508 --> Mouse IgG1 ab102998 --> Rabbit IgG I think that is also what you suggested in the data sheets, I just would like to make sure, that I am correct. Thank you for the assistance. We are in the situation that we - with the exception of the antibody against Ceruloplasmin – don’t have any information whether the antibodies work for the perfusion-fixed, sucrose-immersed, cryostate sectioned brains that we will intend to use these antibodies on. Being responsible for the payment I am therefore a bit hesitant considering the only 5% discount to this relatively large order.
