Recommended dilutions IHC-FoFr: 1/250 (PMID 19186165). IHC-Fr: Use at an assay dependent dilution (PMID 17715346). WB: Use at a concentration of 0.03 - 0.1 µg/ml. Approx 30kDa and 26kDa bands observed in Human Kidney and Human Lung lysates(calculated MW of 30.9kDa, 27.3kDa and 26.4kDa according to NP_000894.1, NP_001020604.1and NP_001020605.1 respecively).
Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

NQO1 is a member of the NAD(P)H dehydrogenase (quinone) family and encodes a cytoplasmic 2-electron reductase. This FAD-binding protein forms homodimers and reduces quinones to hydroquinones. NQO1's enzymatic activity prevents the one electron reduction of quinones that results in the production of radical species. NQO1 functions as an important part of cellular antioxidant defense by detoxifying quinones thus preventing the formation of reactive oxygen species. Mutations in the gene for NQO1 have been associated with tardive dyskinesia (TD), an increased risk of hematotoxicity after exposure to benzene, and susceptibility to various forms of cancer. Altered expression of NQO1 has been seen in many tumors and is also associated with Alzheimer's disease (AD). NQO1 levels have been shown to be raised in may tumour types. This has led to a search for anti-tumour agents that can be activated by NQO1 resulting in specific targetting of these compounds to the tumour site. Many cytotoxic drugs contain the quinone moiety, such as the benzoquinone group, which can be activated by NQO1 through the reduction into hydroquinone [which in this case can auto-oxidize and lead to the formation of free radicals and alkylating species]. However, the rat enzyme has been shown to be significantly more effective in activating some of these compounds than the human and mouse enzymes. This indicates that the available cytotoxic drugs are better substrates for the rat enzyme and are not the most ideal prodrugs for activation by NQO1 in human tumors.
