Question 1

Wednesday 23-May-2012

Dear xxxxx
NQO1 identification in frozen sections of rat brain.

We have tried ab34173 with rather poor and certainly unpublishable results. We wonder if ab2346 would be better, as it seems to work in mice. We have found one paper (attached) in which the authors claim to have used one of your anti-NQO1 antibodies and produced good images in rat brain frozen sections, but they failed to give the catalogue no. I would be very grateful if you were able to find some indication from your records which of the antibodies they used.

With kind regards

ANSWER:

Thank you very much for supplying that information.

I am sorry to hear of your experience with ab34173. As requested, I am trying to track down which antibody was used in the paper you provided. However this is likely to be difficult as we supply our products through distributors in China (where the paper originates from) and I therefore also need their assistance to identify the order. I will however try my best. Unfortunately they haven't even mentioned which secondary antibody they used so I can'tevenreduce the number of possibilities by the species the antibody was raised in.

I have written an email to the author of the paper requesting the information so hopefully they will respond. I'll certainly keep you updated with whatever I find.

I am very sorry that you found ab34173 not to work as expected. I have looked into it and it seems we have had no similar reports and it is quite a popular antibody. It may unfortunately be that you received a faulty vial. Due to the nature of these products this does happen from time to time. If you would like I could provide you an alternative lot of this antibody to try free of charge?

Alternatively, if you would likeI could arrange for the other antibody, ab2346, which we discussed to be sent to you (again freeof charge)? Although as you say the reports where this has been used in frozen sections is primarily with mouse tissue, the antibody has been shown to detect the the rat protein and would be guaranteed by the Abpromise if you were not satisfied with the results. You would then be entitled to your money back or an alternative antibody if the problem is reported within 6 months of purchase.

In order to arrange for either of these options could you please provide me with the following information:

1. The order number (or approximate date of delivery and delivery address) of ab34173

2. A brief description of how the tissue was fixed?

3. What dilution of primary antibody was used and the incubation conditions?

4. What blockingconditions were used?

5. If you could provide an image of the results obtained that would be very helpful.

This information will also allow me to lookinto the problems encountered with ab34173 more fully and see if any additional testing needs to be implemented from our side.

I look forward to receiving your reply.
