Application notes
(see key)
Recommended dilutions ICC/IF: Use at a concentration of 1 µg/ml. WB: Use at a concentration of 1 µg/ml. Detects a band of approximately 70 kDa (predicted molecular weight: 70 kDa).
Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

ab43421 gave a positive signal in the following Human Tissue Lysates: Brain, Testis, Heart and Skeletal muscle.
Cellular localization
Nucleus (nucleoplasm). Localized predominantly in the nucleoplasm and at both the nucleoplasmic and cytoplasmic faces of the nuclear pore complex (shuttles between both).

Relevance
NXF1 (Nuclear Export Factor 1)is a 70 kDa protein involved in the nuclear export of mRNA species bearing retroviral constitutive transport elements (CTE) and is thought to be a principal export receptor for cellular mRNA, from the nucleus to the cytoplasm. NXF1 is primarily located in the nucleoplasm but it can also be found in association with both the cytoplasmic and the nucleoplasmic ends of the nuclear pore complex. It contains an N-terminal nuclear localization and export region (NLS-NES), a central RNA binding domain (RBD), and a C-terminal portion that is required for nuclear localization and nuclear rim association. It is expressed ubiquitously.

Immunocytochemistry/ Immunofluorescence - NXF1 antibody (ab43421)
ICC/IF image of ab43421 stained human HeLa cells. The cells were methanol fixed (5 min), permabilised in PBS-T (20 min) and incubated with the antibody (ab43421, 1µg/ml) for 1h at room temperature. 1%BSA / 10% normal goat serum / 0.3M glycine was used to quench autofluorescence and block non-specific protein-protein interactions. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red). DAPI was used to stain the cell nuclei (blue).
