Neuropeptide S (NPS) is a novel target in the neurobiology of sleep/wakefulness regulation and anxiety-like behavior. NPS potently promotes arousal and suppress all stages of sleep. NPS acts via its cognate receptor (NPSR) and induces mobilization of intracellular Ca2+ stores. NPSR mRNA is widely distributed in the brain, including the amygdala and the midline thalamic nuclei. The NPS peptide precursor is primarily expressed between the locus coeruleus (LC) and Barrington's nucleus, a part of the brain with known function in arousal, stress, and anxiety.
