Recently I have purchased NRP1 antibody from Abcam (product code : ab25998, lot no: GR1127-4) I want to use this for Immunofluorescence study. In the datasheet it is mentioned that one can use the antibody at 5ug/ml conc for this purpose. However, I could not find anywhere the concentration of the supplied product. |

The concentration is 0.5 mg/ml. The antibody is titered to work at specific dilutions. This is why concentration is not mentioned on the datasheet. I have updated the datasheet with dilutions only to reflect this more clearly. For ICC/IF recommended dilution to start at is 1/200. |

What about this antibody or what about neuropilin explains the 28 kDa discrepancy between the predicted molecular weight and the observed molecular weight? |

We used a transfection model using an HA-tagged human Neuropilin 1 sequence. The expressed protein ran near 130 kDa, similar to the observed molecular weight found in other studies (Proc. Natl. Acad. Sci. U.S.A. 97:2573-2578(2000)). |

All lanes : Anti-Neuropilin 1 antibody (ab25998) at 1/500 dilution
Lane 1 : Untransfected COS-7 cells
Lane 2 : COS-7 cells transfected with HA-tagged Neuropilin 1
Predicted band size : 102 kDa
ICC/IF image of ab25998 stained SHSY5Y cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab25998, 5µg/ml) overnight at +4ºC. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
IHC image of ab25998 staining in human heart formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab25998, 1µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.
