Thank you for contacting Abcam Technical Team and for taking the time to provide some useful details of the experiments. I am very sorry to hear that you are having problems with this product. The protocol looks absolutely fine to me and I understand that you had good results previously with different batches. I would like to reassure you that our Abpromise applies to your complaint since you purchased this product within the guarantee period. This means that in the event that a product is not functioning in the applications/species cited on the product data sheet (and the problem has been reported within 6 months of purchase) we will happily offer a credit note/refund to the value of the product purchased. Currently, we do not have new batch in stock so I can't offer you a different batch. However, we have some alternative antibodies in our catalogue (ab83253, ab27526), please take a look at them and let me know how you wish to proceed with your enquiry. I look forward to hearing from you soon. |

Dear technical team, Our customer would like to purchase 11 antibodies from abcam and needs control for each of antibody. The customer will test IHC-P so, control should be slide type. I have searched only one of them as below and would like to ask check the rest of them. |

Thank you very much for contacting us - and sending the enquiry as a table. That clear overview was very helpful :-) As you may know, we have not tested the individual samples for their expression of the mentioned targets. However, according to the literature and other sources such as http://www.proteinatlas.org/ these tissues exhibit high expression of these proteins and thus are suitable controls. However, we do not currently have an image of ab10800, ab16821, ab30483, ab16822, and ab78289 in IHC-P. Therefore, I can offer a discount off a future purchase if you buy one of these antibodies now, use it IHC-P and submit an image of your results to us in the form of an Abreview. The discount would be to the value of 1 free primary antibody each. If you decide to go ahead and purchase this product, please contact our Scientific Support team prior to the purchase for more details. I hope this information is helpful to you. Please do not hesitate to contact me if you need any further advice or information in this regard. |

Does ab27526 Notch1 antibody detect active and non active forms of the protein? |
|||||||||
ANSWER: |
Thank you for your recent telephone enquiry. On further investigation, I can confirm that the immunogen for ab27526 Notch1 antibody is located at C-terminus and therefore should recognize the cytoplasmic domain, the active form that translocates into the nucleus. It should also recognize the full length Notch 1. In Western blotting, it should recognize both 120 kDa (active form) and 300 kDa (full length). I hope this is helpful to you. Should you have an further questions, please do not hesitate to contact us. |

Does this antibody recognize the activated form of Notch1? |
|||||||||
ANSWER: |
Thank you for your enquiry. We have not directly tested this antibody with activated form. However, based on the immunogen sequence, it most likely recognizes activated form. Should you require any other information, please do not hesitate to contact me. |

Staining of Ovarian carcinoma using ab27526
ab27526 staining Notch1 in mouse neural tube by immunohistochemistry (Frozen). Sections were paraformaldhyde fixed prior to blocking in 20% serum for 16 hours at 25°C and then incubated with ab27526 for 16 hours at 25°C. A Cy3 conjugated donkey polyclonal to rabbit Ig was used as the secondary antibody. Blue staining is nuclear staining with DAPI and the specific Notch signal is red.

ab27526 staining Notch 1 in Mouse pancreatic cancer primary cells by ICC/IF (Immunocytochemistry/immunofluorescence). Cells were fixed with acetone/methanol (1:1) and blocked with 10% serum for 1 hour at room temperature. Samples were incubated with primary antibody (1/100 in PBS) for 4 hours. An Alexa Fluor®488-conjugated Donkey anti-rabbit IgG polyclonal (1/1000) was used as the secondary antibody. Nuclei were counterstained with DAPI.

ab27526 staining Notch 1 in Human pancreatic neoplastic lesion tissue sections by Immunohistochemistry (IHC-P - formaldehyde-fixed, paraffin-embedded sections). Tissue was fixed with paraformaldehyde and blocked with 10% serum for 1 hour at room temperature; antigen retrieval was by heat mediation in antigen unmasking solution. Samples were incubated with primary antibody (1/100 in PBS) for 8 hours at 4ºC. A Biotin-conjugated Goat anti-rabbit IgG polyclonal (1/1000) was used as the secondary antibody.
