Hello,   We are currently working on fractioning our protein samples to isolate mitochondria. We purchased your nuclear pore protein complex antibody (ab50008) in order to determine whether our fractionation procedure was successful via western blot. We are a little confused, however, about the bands that we see on our blots. We see very thick bands in all of our samples (lanes 2 and 5 are our mitochondria fractions) that appear around 120kDa and then a second band that would appear to show that our mitochondrial fractions are pure, but around 62kDa (please see attached image). The information sheet cites the size as 106kDa, but some of your other NPP antibodies cite specific bands at 62kDa. Any insight that you might provide as to which bands are the correct size for our purposes (or both?) would be greatly appreciated. I can confirm this antibody is sourced externally. The originator has confirmed that it will recognise several proteins in the nuclear pore complex in various species. These proteins range in size between 62 and 210 kDa. Unfortunately they do not have any information to indicate the proteins that each band corresponds to.

They have performed blotting with HeLa cell extract and find banding between 60 and 150kDa.
