Anti-Ogg1 antibody (ab22766) at 1 µg/ml + HeLa (Human epithelial carcinoma cell line) Nuclear Lysate (ab27251) at 20 µg
Secondary
IRDye 680 Conjugated Goat Anti-Rabbit IgG (H+L) at 1/10000 dilution
Performed under reducing conditions.
Predicted band size : 39 kDa
Observed band size : 37 kDa (why is the actual band size different from the predicted?)
Additional bands at : 70 kDa. We are unsure as to the identity of these extra bands.
ICC/IF image of ab22766 stained human HEK 293 cells. The cells were methanol fixed (5 min), permabilised in 0.1% PBS-Tween (20 min) and incubated with the antibody (ab22766, 5µg/ml) for 1h at room temperature. 1%BSA / 10% normal goat serum / 0.3M glycine was used to block non-specific protein-protein interactions. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red). DAPI was used to stain the cell nuclei (blue). This antibody also gave a positive IF result in HeLa, HepG2 and MCF7 cells.
All lanes : Anti-Ogg1 antibody (ab22766) at 1 µg/ml
Lane 1 : Heart (Mouse) Tissue Lysate
Lane 2 : Heart (Rat) Tissue Lysate
Lysates/proteins at 10 µg per lane.
Secondary
Goat polyclonal to Rabbit IgG - H&L - Pre-Adsorbed (HRP) at 1/3000 dilution
developed using the ECL technique
Performed under reducing conditions.
Predicted band size : 39 kDa
Observed band size : 37 kDa (why is the actual band size different from the predicted?)
Exposure time : 8 minutes
ab22766 staining Ogg1 in Human osteosarcoma tissue sections by IHC-P (Formaldehyde-fixed, Paraffin-embedded sections). Tissue samples were fixed with formaldehyde and blocked with 5 minutes of peroxidase followed by 10 minutes of protein at 20ºC; antigen retrieval was by heat mediation in target retrieval solution. The sample was incubated with primary antibody (1/500 in antibody diluent) at 20ºC for 45 minutes. An undiluted HRP-conjugated polyclonal to rabbit IgG was used as secondary antibody.
