Hi,

I have aliquoted my antibody (The cat numbers:ab81093 and ab68779) but
I dont use glycerol that was recommended in datasheet and stored
them for several months in -20°C in freezer and I haven't tested the
antibody to check to see if it is still viable.please help me what I
should do? Can I consolidate all aliquots into the original vial, add
glycerol, and realiquot or add the scaled down recommended amount of
glycerol to each aliquot.Thank you very much.

Thank you for your enquiry.

As glycerol is a cryoprotectant, if the antibody has been frozen for a few months without it, it may regrettablyalready be damaged.

However, I would suggest to try one aliquot of each antibody in an experiment,there is a chance they are still viable and they could work. It may be that a higher concentration is required in case it is degraded.

If they do work, then I would suggest to add 40% (ab68779) or 50% (ab81093) glycerol to each ofthe rest of the aliquots in order to help keep the antibody stable during further storage. For example, if you have a 10 ul aliquot, add 8 or 10 ul of glycerol. As the glycerol does not freeze solid, it should mix slowly with the aliquot of antibody. I would suggest to give the tubes a good mix.

Also I want to know that when you recommend to add glycerol to a
final volume of 50% what you mean?you mean that I should add glycerol
as equal of final volume of ab (I mean if I have 100µl ab I should add
100µl glycerol) or I should add glycerol as 50% of final volume of ab
(I mean if I have 100µl ab I should add 50µl glycerol).Thanks in

When you add glycerol to a final volume of 50%, it means that half of the total finalvolume should be antibody and half of the volume should be glycerol. Therefore, if the antibody is 100ul, you should add 100ul of glycerol. Then you'll have a final volume of 200ul, made of 100ul antibody and 100ul glycerol.

Thank you for contacting us. As discussed over the phone the recommended storage conditions for this antibody (ab81093) is for short term at +4°C (1-2 weeks). For longer term storage we would recommend adding glycerol to a final volume of 50%, aliquoting then storing at -20°C. Freeze/thaw cycles should be avoided.
