    Applications

    Application notes

This antibody gave a positive result in ELISA against the immunizing peptide (ab40904).
Not yet tested in other applications.

Optimal dilutions/concentrations should be determined by the end user.

    Target

    Function

Appears to modulate osteoblastic differentiation. Could also function as an autocrine and paracrine factor linked to glucose metabolism in skeletal muscle.

    Tissue specificity

    Expressed in bone.

    Cellular localization

    Secreted.

    All lanes : Anti-Osteocrin antibody (ab40905) at 1 µg/ml

Lane 1 : Skeletal Muscle (Rat) Tissue Lysate - normal tissue (ab29376)
Lane 2 : Liver (Rat) Tissue Lysate
Lane 3 : HeLa (Human epithelial carcinoma cell line) Whole Cell Lysate
Lane 4 : HepG2 (Human hepatocellular liver carcinoma cell line) Whole Cell Lysate
Lane 5 : Skeletal Muscle (Human) Tissue Lysate - adult normal tissue (ab29330)
Lane 6 : Thymus (Human) Tissue Lysate - adult normal tissue (ab30146)

Lysates/proteins at 10 µg per lane.

    Secondary
Goat polyclonal to Rabbit IgG - H&L - Pre-Adsorbed (HRP) at 1/3000 dilution

    Performed under reducing conditions.

Predicted band size : 15 kDa
Observed band size : 50 kDa (why is the actual band size different from the predicted?)

Osteocrin has previously been shown to run at 5 & 11 kDa, with the MW of the unprocessed precursor being 15 kDa. We are unsure as to why ab49405 is giving a MW of 50 kDa. For Rat, however, SwissProt suggests that expression of Osteocrin is almost exclusively seen in Skeletal Muscle. Abcam welcomes customer feedback and would appreciate any comments regarding this product and the data presented above.
