Recommended dilutions ELISA: Use at a concentration of 0.5-1 µg/ml (indirect method). ICC/IF: 1/50. (Based on Abreview.) Neut: Use at a concentration of 1-3 µg/ml. WB: Use at a concentration of 0.1-0.2 µg/ml. Detects a doublet band of approximately 65 kDa and a 30 kDa band (due to glycosylation).
The Neutralization Dose50 (ND50) for anti-mouse osteopontin is approximately 1 to 3 µg/ml in the presence of 2 µg/ml of recombinant mouse osteopontin using 293 cells.
Not tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Osteopontin is the principal phosphorylated glycoprotein of bone and is expressed in a limited number of other tissues including dentine. Osteopontin is produced by osteoblasts under stimulation by calcitriol and binds tightly to hydroxyapatite. It is also involved in the anchoring of osteoclasts to the mineral of bone matrix via the vitronectin receptor, which has specificity for osteopontin. Osteopontin is overexpressed in a variety of cancers, including lung, breast, colorectal, stomach, ovarian, melanoma and mesothelioma.
