Recommended dilutions ChIP: Use at an assay dependent dilution (PMID 18057103). ICC/IF: Use at a concentration of 0.8 - 4 µg/ml. IHC-Fr: Use at a concentration of 0.8 µg/ml. WB: Use at a concentration of 1 µg/ml. Detects a band of approximately 37 kDa (predicted molecular weight: 37 kDa). Can be blocked with Otx2 peptide (ab24347).
Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Homeobox protein Otx2 is a member of the bicoid sub-family of homeodomain-containing transcription factors. It is an early marker of endodermal differentiation of embryonic stem cells and may play a role in the development of the brain and the sense organs. Two distinct isoforms have been identified. Can bind to the BCD target sequence (BTS): 5'-TCTAATCCC-3'.

All lanes : Otx2 antibody - ChIP Grade (ab21990) at 1 µg/ml
Lane 1 : Y79 Lysate Lane 2 : Y79 Lysate with Otx2 peptide (ab24347) at 1 µg/ml
Lysates/proteins at 20 µg per lane.
Secondary Alexa Fluor Goat polyclonal to Rabbit IgG at 1/10000 dilution
Predicted band size : 37 kDa Observed band size : 37 kDa Additional bands at : 28 kDa (possible cleavage fragment,isoform,non-specific binding).
ab21990 recognised a band corresponding to the predicted size of Otx2 in Y79 cell lysate (lane 1, see arrow). The band was blocked using the immunizing peptide (lane 2). A less intense band was observed at ~28 kDa, which may be a cleavage product or an Otx2 isoform. Alternatively the ~28 kDa band may respresent non-specific binding by the antibody.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Otx2 antibody - ChIP Grade (ab21990)
Mouse E6 embryos were sectioned and then fixed in paraformaldehyde. Antigen retrieval was performed using citric acid and the sections permeablized and blocked for 1 hour in serum. Embryos were stained with ab21990 (1/2500) diluted in 10% serum, 1% ovalbumin in PBSTween for 14 hours at 4°C. They were then washed and stained with a goat anti-rabbit biotin conjugated antibody. After using a biotinylated secondary, ABC and then DAB was used for color development.
