All lanes : Anti-P4HA2 antibody (ab70887) at 0.04 µg/ml
Lane 1 : HeLa whole cell lysate at 50 µg
Lane 2 : HeLa whole cell lysate at 15 µg
Lane 3 : HeLa whole cell lysate at 5 µg
Lane 4 : 293T whole cell lysate at 50 µg
developed using the ECL technique
Predicted band size : 61 kDa
Observed band size : 61 kDa
Additional bands at : 28 kDa,33 kDa,50 kDa. We are unsure as to the identity of these extra bands.
Exposure time : 3 minutes
HeLa whole cell lysate (at 1 mg for IP, 20% of IP loaded) with ab70887 at 1 µg/ml for WB subsequent to IP at 3 µg/mg lysate.
Lane 2: control IgG.
Detection: Chemiluminescence with an exposure time of 30 seconds.
All lanes : Anti-P4HA2 antibody (ab70887) at 1 µg/ml
Lane 1 : Whole cell lysate prepared from 9L cells
Lane 2 : Whole cell lysate prepared from PC12 cells
Lane 3 : Whole cell lysate prepared from RIE-1 cells
Lysates/proteins at 20 µg per lane.
Secondary
HRP conjugated pig anti-rabbit polyclonal at 1/5000 dilution
developed using the ECL technique
Predicted band size : 61 kDa
Observed band size : 61 kDa
Additional bands at : 35 kDa (possible degradation product).
Exposure time : 5 minutes
Good immunoreactivity observed in RIE-1 cell lysate that is consistent with the mobility of full-length P4HA2. A faster migrating species of ~35 kDa has been commonly observed with other antibodies (including this one) that could be a degradation product or alternatively spliced form of P4HA2. Further charcaterisation is required.
