Relevance

PACS1 is a coat protein that is involved in the localization of trans Golgi network (TGN) membrane proteins that contain acidic cluster sorting motifs. In addition, the human protein plays a role in HIV-1 Nef-mediated downregulation of cell surface MHC-I molecules to the TGN, thereby enabling HIV-1 to escape immune surveillance. PACS1 is known to interact with HIV-1 Nef, AP-1 and AP-3 but not with AP-2.

ab56072 used in Western Blot.

Approximately 2 × 106 (HeLa or Jurkat) cells were disrupted in 0.5 ml of lysis buffer, and initially 50 μl of 15,000 × g supernatants were immunoblotted for actin. After adjusting the volume to constant actin values, the indicated vesicular or adapter proteins were detected by immunoblotting extracts of cells treated with the siRNAs listed below the respective blots.
