Recommended dilutions WB: Use at an assay dependant dilution. Use a lysis buffer made with Tris HCl (pH 8.1) and 1%laurylmaltoside (with common protease inhibitors). Use under non-reducing conditions. Csk binding protein is an ubiquitously expressed 46 kDa transmembrane adaptor protein present in membrane microdomains (rafts), which however migrates on SDS-PAGE gels anomalously as an 80 kDa molecule. The reason is, that PAG has a pI value of about 4.6 and binds poorely SDS and therefore it had decreased mobility on SDS gels.
Is unsuitable for IP (use ab4206 or ab14989).
May be unsuitable for ICC-IF due to unsuccesful results in submitted Abreview. (Please see Abreview comments)
Not tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Csk binding protein, also known as PAG (phosphoprotein associated with GEMs), is a ubiquitously expressed 46 kDa transmembrane adaptor protein present in membrane rafts (glycosphingolipid-enriched microdomains), which however migrates on SDS PAGE gels anomalously as an 80 kDa molecule. Following tyrosine phosphorylation by Src family kinases, PAG binds and thereby activates the protein tyrosine kinase Csk, the major negative regulator of the Src family kinases. Signaling via the B-cell receptor in B cells or high affinity IgE receptor (FcepsilonRI) in mast cells leads to PAG increased tyrosine phosphorylation and Csk binding, while T cell receptor signaling causes PAG dephosphorylation, loss of Csk binding and increased activation of the protein tyrosine kinase Lck.

Immunoprecipitation
1. Frozen Raji cells lysate, immunoprecipitated with ab4206

Reprobe with ab9527.
The immunoprecipitation used covalently bound antibody to Sepharose beads as immunosorbent. This method of precipitation obviously differs from proteinA/G-sepharose or GoatxMs -Sepharose mediated immunoprecipitation protocols.
