We have injected the homogenized tumor tissue isolated from human lung into the armpit of mice. After certain time the neoplasm is formed in the armpit. Now we want to know the nature of the neoplasm that is originally from human lung tumor or is a new tumor from mouse. We would like to check the nature of the neoplasm by IHC (florescence). Could you please tell us which antibody that can specifically recognize human tissue (tumor) or mouse tissue (tumor) ? We hope that antibody does not cross-react on human or mouse. Also, the host of antibody is not form mouse. |

Thank you for contacting us. I have heard of people using anti-vimentin clone V9, to detect human tissue implanted in mice. However, this is a mouse monoclonal and our version, ab8069 is unconjugated. We do not have a version conjugated to a fluorochrome, enzyme, or other detector. An anti-mouse IgG secondary may give some staining of endogenous IgG, as you are probably aware. For detecting mouse tissue without detecting human, consider ab15630, with is also available biotinylated as ab15631 and FITC-conjugated as ab15638: http://www.abcam.com/index.html?datasheet=15630 The ab15630 and ab15631 have been shown to work on frozen tissue, but will not work on paraffin-embedded tissue. Ab15638 has only been tested for flow cytometry so we canot guarantee it will work for IHC. Antibody ab6022 has not been tested on mouse tissue. An alignment of the immunogenic peptide with the mouse protein shows that it is 69% identical; it is difficult to predict from this if the antibody will or will not cross-react with the mouse protein. Ab6022 (10ug/ml) staining human PAR6 in human pancreas by immunohistochemistry using paraffin embedded tissue. Microwaved antigen retrieval with Tris/EDTA buffer pH9, HRP-staining.
Ab6022 shows strong cytoplasmic staining in rare cells of intralobular ducts.
ab6022 at 5ug/ml staining PAR6 in human heart tissue section by Immunohistochemistry (Formalin/PFA fixed paraffin-embedded sections). Tissue underwent antigen retrieval in steam with citrate buffer (pH 6.0). The AP-staining procedure was used for detection.
