This antibody recognises the peptide enzyme designated proprotein convertase 1 and also known as PC1/3, NEC1 and neuroendocrine convertase 1. This enzyme is involved in the processing of hormone (and other protein) precursors including renin, somatostatin and insulin. It is found in secretory granules. The PC1/3 antibody recognises the endogenous protein in rat and human pancreatic islets and in the RIN cell line. We have also seen reactivity in brain extracts though this is faint unless glycoproteins are enriched by affinity-purification on concanavalin A. The protein recognized by this antibody has 3 forms. These proteins are a 93 kDa which is the full length glycosylated protein (i.e. includes the signal peptide), a 91 kDa which is the glycosylated protein less the signal peptide, and a 66 kDa which is the active protein. In WB, this antibody only picks up a 66 kDa band. The 93 kDa protein is unlikely to occur in nature due to the removal of the signal peptide during co-translational insertion into the endoplasmic reticulum, and the 91 kDa protein may occur briefly during transport from the ER to the Golgi, where the propeptide is removed to yield the active 66 kDa protein. Since these processing steps all occur at the N-terminus and the antibody is directed at the C-terminus, the antibody would be expected to recognize all three forms.

Involved in the processing of hormone and other protein precursors at sites comprised of pairs of basic amino acid residues. Substrates include POMC, renin, enkephalin, dynorphin, somatostatin and insulin.
Defects in PCSK1 are the cause of proprotein convertase 1 deficiency (PC1 deficiency) [MIM:600955]. PC1 deficiency is characterized by obesity, hypogonadism, hypoadrenalism, reactive hypoglycemia as well as marked small-intestinal absorptive dysfunction It is due to impaired processing of prohormones.
Belongs to the peptidase S8 family. Furin subfamily.
