Phoned to ask for products to test for PCSK2 in ELISA. |
ANSWER: |
|
Thank you for contacting us.
The Rabbit polyclonal to PCSK9 (ab125251;http://www.abcam.com/PCSK9-antibody-ab125251.html) which we spoke about earlier is our only product that has been tested in ELISA. It recognizes a region around aa100-300 of Human PCSK9. The process which we were speaking of would be a sandwich ELISA.This product has not been tested in sandwich ELISA, or sELISA on our web sheet, which is a different application on our site. Sandwich ELISA procedures can be difficult to optimize and tested match pair antibodies should be used. This ensures the antibodies are detecting different epitopes on the target protein so they do not interfere with the other antibody binding. Therefore, we are unable to guarantee our antibodies in sandwich ELISA unless they have been specifically tested for sandwich ELISA.
If you were to use this product in an indirect or direct ELISA it would be guaranteed. A great secondary for use in that system would beGoat polyclonal Secondary Antibody to Rabbit IgG - H&L (Biotin), pre-adsorbed (ab97067;http://www.abcam.com/Goat-polyclonal-Secondary-Antibody-to-Rabbit-IgG-H-L-Biotin-pre-adsorbed-ab97067.html). This product is Biotinylated and preabsorbed so you should experience minimal background with it.
A positive control protein for this would bePCSK9 peptide Propeptide domain (ab44562,http://www.abcam.com/PCSK9-peptide-Propeptide-domain-ab44562.html).
However, the advantage of sandwich ELISA is that the sample does not have to be purified before analysis, and the assay can be very sensitive (up to 2 to 5 times more sensitive than direct or indirect). The problem is in choosing two antibodies that will not interfere with each other. If you wish to perform this assay I would recommend the following products as they are derived from different regions of the protein:
Rabbit polyclonal to PCSK9 (ab125251;http://www.abcam.com/PCSK9-antibody-ab125251.html)andGoat polyclonal to PCSK9 (ab28770;http://www.abcam.com/PCSK9-antibody-ab28770.html). These are against different regions on the protein and should interfere with one another.
You will also need to biotinylate one of these so that you will be able to use the streptavidin detection system that you spoke of. I would suggest using our EasyLink conjugation kit for Biotin Type A (ab102865;http://www.abcam.com/EasyLink-Biotin-type-A-Conjugation-Kit-3-x-10%C2%B5g-Biotin-ab102865.html).
We do not currently offer PCSK9 proteins which span the regions needed for both of these antibodies. You may be able to find this using Biocompare.com, a Life Sciences search engine which scans all the Life Science companies.
I hope this information is helpful to you. Please do not hesitate to contact us if you need any more advice or information. |
