  • Product namePGK1 protein (Tagged-His Tag)See all PGK1 proteins and peptides ...
  • Protein descriptionRecombinant fragment, corresponding to amino acids 1-409 of Human PGK1 with N terminal His tag. MWt 47kDa; UniProt ID = P00558
  • Expression hostE. coli
  • Properties

  • Purification notesPurity level >85%
This protein was expressed as an N-terminal His-tag fusion protein using Escherichia coli, and purified using Immobilized Metal Ion Affinity Chromatography. In some cases, smaller protein fragments may be present in addition to the intended expression product as a result of premature termination during translation in E. coli and subsequent co-purification via the His-tag. In some cases purified proteins run at a molecular weight different to the theoretically calculated molecular weight. This may be as a result of unequally distributed charges in the amino acid sequence. Alternatively, dimerisation of the expression product can occur under oxygen limitation during expression/cultivation.

  • FunctionIn addition to its role as a glycolytic enzyme, it seems that PGK-1 acts as a polymerase alpha cofactor protein (primer recognition protein).
  • PathwayCarbohydrate degradation; glycolysis; pyruvate from D-glyceraldehyde 3-phosphate: step 2/5.
  • Involvement in diseaseDefects in PGK1 are the cause of phosphoglycerate kinase 1 deficiency (PGK1D) [MIM:300653]. It is a condition with a highly variable clinical phenotype that includes hemolytic anemia, rhabdomyolysis, myopathy and neurologic involvement. Patients can express one or more of these manifestations.
  • Sequence similaritiesBelongs to the phosphoglycerate kinase family.
  • Cellular localizationCytoplasm.
