Recommended dilutions ELISA: 1/1000. ICC/IF: Use at a concentration of 1 µg/ml. IHC-P: 1/50 - 1/100. WB: 1/100 - 1/500. Detects a band of approximately 58 kDa (predicted molecular weight: 58 kDa).
Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Cytoplasm. Nucleus. Translocates to the nucleus in response to different apoptotic stimuli. Nuclear translocation is sufficient to induce cell death that is caspase independent, isoform specific and independent of its enzymatic actvity.

PKM2 (pyruvate kinase, muscle) catalyzes the production of phosphoenolpyruvate from pyruvate and ATP. PKM2 has been shown to interact with thyroid hormone, and thus may mediate cellular metabolic effects induced by thyroid hormones. It has been found to bind Opa protein, a bacterial outer membrane protein involved in gonococcal adherence to and invasion of human cells, suggesting a role of this protein in bacterial pathogenesis. Three alternatively spliced transcript variants encoding two distinct isoforms have been reported.
There are 4 isozymes of pyruvate kinase in mammals: L, R, M1 and M2. Ab38237 recognizes the M1/M2 isoform of pyruvate kinase

ab38237, at 1/50 dilution, staining PKM2 in Human breast carcinoma by Immunohistochemistry, Formalin-fixed, Paraffin-embedded tissue. ab38237 was peroxidase-conjugated to the secondary antibody, followed by AEC staining.

Immunocytochemistry/ Immunofluorescence-PKM2 antibody(ab38237)
ICC/IF image of ab38237 stained HeLa cells. The cells were 4% PFA fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab38237, 1µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
