Question 1

Thursday 17-June-2010

I'd like to detect human PON1 protein expressed in the liver. But PON2 and PON3 are also expressed in the liver, so I need to use a PON1 specific antibody that don't cross-react to PON2 or PON3. Do you habe any information on the cross-reactibity of your 6 anti-PON1 antibodies with PON2 and PON3? Which product do you recommend for my purpose? Thank you.

ANSWER:

Thank you for your patience.

I have received some confirmation about some products and would like to update you on the information.

ab85347: We have not tested this product for cross reactivity with PON2 and PON3.

ab40775: We have not tested this product for cross reactivity with PON2 and PON3.

ab53193: This antibody has been raised to a synthetic peptide corresponding to amino acids 298-309 of human PON1, conjugated to KLH via an N-terminal cysteine residue. The corresponding sequence differs by one amino acid in mouse and two amino acids in rat. The peptide sequence differs by one amino acid in human PON2 and two amino acids in human PON3. Due to the homology between these two sequence, it is likely that some cross reactivity with PON2 and PON3 may occur. A 1 amino acid difference in the peptide would translate to 90% and 2 amino acid difference would be closer to 80% homology. Thus there is a less chance that PON3 would cross react.

ab92466: We have not tested this product but there is 57.1% homology with PON2 (Q15165) and 50.0% identity with PON3. Therefore, we do not expect cross-reactivity with either one.

I am still waiting for information on ab24261 and ab26930 and will be able to get back to you once I receive confirmation from the originator.

Please bear in mind that all these products were not tested for cross reactivity with PON2 nor PON3. Therefore, we really do not know for sure.

However, I can reassure you that all of our products are guaranteed to work in species and application as stated on the datasheet. Should you not get expected results, then please inform us within the guarantee period of 120 days (Abpromise). We will help troubleshoot the experiment and if the product is indeed faulty, we will be more than happy to offer a free replacement or refund.

I hope our Abpromise guarantee will give you the assurance you need to use our products in your research.
