Specificity 

ab64346 recognizes human eosinophil major basic protein irrespective of the stages of eosinophil activation. It recognizes the 28.3kD secreted pro-form of MBP (Pregnancy Associated MBP) found in the placenta and serum of pregnant women as well as the 13.8kD mature form of the protein which is found in the matrix of the eosinophil large specific granual.

Recommended dilutions
IHC-P: Use at an assay dependent dilution. Perform enzymatic antigen retrieval before commencing with IHC staining protocol.
IHC-Fr: Use at an assay dependent dilution. Tissues were fixed in 1:1 solution of methanol and acetone.
Note: The antibody reacts better in frozen tissues than in paraffin embedded tissues.

WB: Use at an assay dependent dilution. Purified normal human eosinophils were treated with digitonin (5mg/ml) for 10 minutes before boiling in sample buffer. Predicted molecular weight: 25 kDa. Purified MBP migrates at 14kD, with typical human samples ranging from 10-16kD.


Not yet tested in other applications.
Optimal dilutions/concentrations should be determined by the end user.

Secreted. Note=The proform is secreted. The mature protein is found in the matrix of the eosinophil's large specific granule (crystalloid core).

Relevance 

PRG2 is a cytotoxin and helminthotoxin. It also induces noncytolytic histamine release from human basophils and is involved in antiparasitic defense mechanisms and immune hypersensitivity reactions. The proform acts as a proteinase inhibitor, reducing the activity of PAPPA. In pregnancy serum, the proform exists as a disulfide-linked 2:2 heterotetramer with PAPPA, as a disulfide-linked 2:2 heterotetramer with AGT, and as a complex (probably a 2:2:2 heterohexamer) with AGT and C3dg.
