Recommended dilutions IHC-FrFl: 1/10 (PMID 19228947) IHC-P: Use at a concentration of 10 µg/ml. WB: Use at a concentration of 0.3 - 1 µg/ml. Detects a band of approximately ~105 kDa (predicted molecular weight: ~105 kDa). Can be blocked with PSD95 peptide (ab12419) or PSD95 peptide (ab12419).
Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Relevance
|
PSD95 is a member of the membrane-associated guanylate kinase (MAGUK) family. It heteromultimerizes with DLG2. With DLG2 it is recruited into the same NMDA receptor and potassium channel clusters. These two MAGUK proteins may interact at postsynaptic sites to form a multimeric scaffold for the clustering of receptors, ion channels, and associated signaling proteins.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - PSD95 antibody (ab12093)
ab12093 PSD95 antibody immunostaining brain tissue (4% PFA perfused paraffin embedded 5 micron saggital section). ab12093 was used at 10 µg/ml for a 20 minute incubation at 37 degrees C (on a Ventana Discovery slide processor). Light antigen retrieval and then light protease treatment is recommended before addition of primary antibody ab12093 in this paradigm. Chris Anderson, Wellcome Trust Sanger Institute, United Kingdom
