IHC-FrFl: Use at an assay dependent concentration. (PubMed: 19228947)
WB: Use a concentration of 0.3 - 1 µg/ml.Detects a band of approximately 105 kDa (predicted molecular weight: 105 kDa).Can be blocked with PSD95 peptide (ab12419).
IHC-P: Use a concentration of 10 µg/ml
Interacts with the cytoplasmic tail of NMDA receptor subunits and shaker-type potassium channels. Required for synaptic plasticity associated with NMDA receptor signaling. Overexpression or depletion of DLG4 changes the ratio of excitatory to inhibitory synapses in hippocampal neurons. May reduce the amplitude of ACCN3 acid-evoked currents by retaining the channel intracellularly. May regulate the intracellular trafficking of ADR1B.

Palmitoylation of isoform 1 is required for targeting to postsynaptic density.
Cell membrane. Cell junction > synapse > postsynaptic cell membrane > postsynaptic density. Cell junction > synapse. Cell junction > synapse > synaptosome. High levels in postsynaptic density of neurons in the forebrain. Also in presynaptic region of inhibitory synapses formed by cerebellar basket cells on axon hillocks of Purkinje cells.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - PSD95 antibody (ab12093)

ab12093 PSD95 antibody immunostaining brain tissue (4% PFA perfused paraffin embedded 5 micron saggital section). ab12093 was used at 10 µg/ml for a 20 minute incubation at 37 degrees C (on a Ventana Discovery slide processor). Light antigen retrieval and then light protease treatment is recommended before addition of primary antibody ab12093 in this paradigm.
