Thanks a lot for your detail information, which is very helpful. I have considered all product you suggested based on my plan. I think I will buy the  Pan-specific phosphoserine antibody  (ab9332) as you mentioned. The first application is that I will use it to detect a purified protein phosphorylation by GSK3b after kinase assay by  western blot. The purified protein is mouse origin. If it works well, I will apply this antibodies in other experiments in vivo by using immunostaining in different cell line and tissues. The cells and tissues come from mouse,rat, monkey and human. Is there any new application there? I will order the antibodies today. What's the best price for that?

Thanks for your information. It is glad to hear that it is Pan-specific phosphoserine antibody. I am going to use it to detect the phosphorylation of purified recombinant dynein intermediate chain of  mouse origin by a Ser/Thr kinase. Although I am not sure it's serine phosphorylation, it has higher possibility than Thr phosphorylation. Therefore, I am wondering if you could help me check if Abcam has  pan-specific phospho-Ser/Thr  antibody. Otherwise, I am going to get  this Pan-specific phosphoserine antibody and pan-specific phosphothreonine antibody(if there is any). if our research is qualified for discount, please let me know. While not truly Pan phospho Ser/ThrAb14764 detects phosphoserine or phosphothreonine in the context of tyrosine, tryptophan or phenylalanine at the -1 position or phenylalanine at the +1 position. This product has been tested and guaranteed to work inICC/IF,ELISA,IP,and WB and has been referenced in seven publications to our knowledge. The product page is:

Ab117253detects free and conjugated Phosphoserine/threonine as well as their modified cellular proteins. It does not cross-react with nonphosphorylated tyrosine, serine, threonine, AMP, or ATP. 1. Dose this antibody recognize all kinds of phospho-serine sites or only recognize the phospho-serine site with certain amino acids around it? 2. May I buy and use a little amount of sample to test my protein first? If it works well, we will buy more. Thanks

Thank you for contacting us.The phosphoserine antibody is pan-specific, however, steric hindrance by some adjacent amino acids may prevent efficient antibody recognition. We have not performed extensive studies to further characterize the affect of surrounding amino acids.

I am trying to identify the kinase that phosphorylates a protein of my interest and would like to use your phosphoserine antibodies. But I am not sure if your antibody works regardless of the surrounding residues.

We still have problem with the antibodies. We are now working on two antibodies which has beengenerouslysent from your company to replace the old vial we have purchased before. One is Anti-Phosphoserine( ab9332 ), the other is Anti-phosphoserien/threonine/tyrosine antibody (SPM101)(ab15556). Unfortunately,none of them could detect any bands for our IP samples. Quite often, the membranes are too dirty to see any bands.
By carefully going through the Western blotting of phospho-proteins protocol on Abcam website, we suspected two possible reasons that we might ignore before.
The first one is we load the samples on NuPAGE 10% Bis-Tris Gels(Novex life technologies) and transfer the membrane on iblot Gel transfer stacks nitrocellulose, regular (Invitrogen). It is different from what you recommend of using PVDF membrane. Do you think by using nitrocellulose membrane, the sensitivity of the membrane will be decreased?
The second one we did not filter the blocking media as you recommend before use. Could you explain a bit more about what to use and how to filter the 5%BSA?
