Question 3

Tuesday 27-September-2005

Dear Dr. Spotswood, With regards to ab16006 (against prealbumin), I was wondering if you could provide more info. I need to know how much of the antibody I need to purchase in order for me to run approximately 100 plasma samples. I appreciate your time and will be waiting for your reply.

ANSWER:

Thank you for your enquiry.

I do not have specifics for the application of ab16006 by sandwich ELISA. However, a general recommendation is to apply the antisera at 20 µg/mL in PBS. Given that this should be applied at 50ul per well and that a vial of ab16006 is sold as 250ul of 0.45 mg/ml. By my calculations this equates to 112.5ug and will allow you to prepare 5.625ml of antiserum dilution and experiment on 112 (112.5) wells.

However, this does not account for development/ optimisation steps that you will have to do and failures. Also 20 µg/mL is a general figure that will depend on the titre of the antibody.

Please do not hesitate to contact me should you require further assistance.
