Our Abpromise guarantee covers the use of ab50667 in the following tested applications.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
ICC/IF: Use a concentration of 10 - 20 µg/ml.
Flow Cyt: 1/200((see Abreview))
WB: Use a concentration of 1 - 2 µg/ml.Detects a band of approximately 15 kDa (predicted molecular weight: 15 kDa).
IP: Use at an assay dependent dilution.
Binds to actin and affects the structure of the cytoskeleton. At high concentrations, profilin prevents the polymerization of actin, whereas it enhances it at low concentrations. By binding to PIP2, it inhibits the formation of IP3 and DG.
Belongs to the profilin family.
Cytoplasm > cytoskeleton.

Immunoprecipitation - Profilin 1 antibody - Carboxyterminal end (ab50667)

Lane 1: Positve control — Western blot: ab50667 at a concentration of 1μg/ml on whole extract of Mouse 3T3 cells at 10μg/ml.
Lane 2: Immunoprecipitation — 10 μg of ab50667 was used to immunoprecipitate Mouse Profilin 1 from 3T3 cell lysate (20μg/ml). Detection antibody: Rabbit Anti-Profilin 1 (N-terminal) antibody.
Lane 3: Negative control — non-relevant antibody was used for IP. Detection antibody: Rabbit Anti-Profilin 1 (N-terminal) antibody
Flow Cytometry - Profilin 1 antibody - Carboxyterminal end (ab50667)

ab50667 detecting Profilin 1 in Human platelets by Flow Cytometry. Platelets were isolated by spinning Platelet rich plasma on Histopaque. Cells were fixed in PFA and permeabilized in 0.1% Triton-X100 in 2% BSA for 30 minutes. The cell population was gated on platelets. The primary antibody was diluted 1/200 in 2% BSA in PBS and incubated with the sample for 18 hours at 4°C. An Alexa Fluor® 488-conjugated Chicken anti-Rabbit IgG (H+L) antibody, diluted 1/500 was used as the secondary antibody.
Abbreviations in the figure:
US Unstained, Red Peak;
IgG Rb: IgG Rabbit (Isotype Control), Blue Peak;
Profilin: Profilin 1, Green peak.

Western blot - Anti-Profilin 1 antibody - Carboxyterminal end (ab50667)

Anti-Profilin 1 antibody - Carboxyterminal end (ab50667) at 1/1000 dilution + Human platelet lysate at 20 µg
Secondary
HRP-conjugated goat anti-rabbit polyclonal at 1/10000 dilution
developed using the ECL technique
Performed under reducing conditions.
Predicted band size : 15 kDa
Observed band size : 17 kDa (why is the actual band size different from the predicted?)
Exposure time : 1 minute
