Thank you for your email. As a positive control I can recommend to use sections from breast, placenta, kidney, liveror pancreas. Indeed, the prolactin receptor is expressed in those tissues. (http://www.uniprot.org/uniprot/P16471) There should however also be an expression in the salivary gland, as seen on the datasheet of ab2772. We do have in our catalogue tissue slides which can be used as positive controls. Our order number xxx We have problem about Positive control, we use gland parotis tissue for Positive control and we used method IHC-P, but we can't found Positive Result. We take product complaints very seriously, and investigate every product that we feel may not be performing correctly. Our products are covered by our guarantee. We will therefore look together through the protocol, and try to find a solution to the problem. If the vial would be faulty, we would replace it for you. As shown on the Abreviews (feedback obtained from end users), the prolactin receptor antibody ab2772 has been successfully used in IHC-Fr on Mouse skin sections and in IHC-P on Human salivary gland sections fixed with paraformaldehyde : http://www.abcam.com/index.html?pageconfig=reviews&intAbID=2772 .
For IHC-P, the antibody was used at a dilution 1/100, for 12 hours at 4°C. No antigen retrieval method was applied.
Another positive control recommended for alternative prolactin receptor antibodies is breast carcinoma.

ab2772 staining the Prolactin receptor in Human salivary gland tissue sections by Immunohistochemistry (IHC-P - paraformaldehyde-fixed, paraffin-embedded sections). Tissue was fixed with paraformaldehyde and blocked with 10% serum for 1 hour at 20ºC; antigen retrieval was not performed. Samples were incubated with primary antibody (1/100) for 12 hours at 4ºC. A Biotin-conjugated Rabbit anti-mouse polyclonal (1/200) was used as the secondary antibody.

ab2772 staining the prolactin receptor in Mouse skin tissue sections by Immunohistochemistry (IHC-Fr - frozen sections). Tissue was fixed with methanol, permeabilized with 0.25% Triton X and blocked with 15% serum and mouse IgG for 1 hour at 25ºC. Samples were incubated with primary antibody (1/20) for 16 hours at 4ºC. An Alexa Fluor®488-conjugated Goat anti-mouse polyclonal (1/250) was used as the secondary antibody.

Overlay histogram showing MCF-7 cells stained with ab2772 (red line). The cells were fixed with 80% methanol (5 min) and incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab2772, 2µg/1x106 cells) for 30 min at 22ºC. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (
