Hello,
Thank you for your answer. I understand your concern about the possible non-specific signal. However, we usually use other mouse antibodies on prostate IHC of mouse and we hardly see any non-specific signal and always localized to the extracellular matrix surrounding the glands and/or the lumen. We don't see non-specific signal in the epithelium. So, my question is: will it be ok if I try the antibody in our usual conditions or do you require I use the mouse blocking system to get the discount?

Hello,
I
would like to test the Anti-Prolactin Receptor antibody [U5] (ab2772)
in IHC of paraffin-embedded mouse prostate tissue. According to the
datasheet, this antibody recognizes rat, rabbit, human, and pig PRLR,
but it hasn't been tested on mouse. Could you let me know if I would get
the 100% testing discount if I do the test and submit the abreview?
Also, I welcome any suggestions for the tests that might not be included
in the datasheets. Or suggestions of other antibodies that might be more
suitable for my needs.

Thank you for your enquiry and your interest in our product.

We have already offered a testing discount for one of our customers who has tested ab2772 on mouse skin tissue sections. The results have been published as an Abreview and the image can be seen on the on-line datasheet. Since the rating of this feedback was only 3 star (and not 5 star), we are not convinced that ab2772 is suitable for detecting murine Prolactin Receptor antibody and have not updated the tested species section with mouse. We would still welcome more results from other customers.

Looking at this particular image, it seems that the background signal is pretty high. Since this primary antibody is raised in mouse and the samples are from the same species, it is likely that the antibody may recognize some endogenous non-specific immunoglobulins.

I could certainly offer you a discount code for testing but it would be worth perhaps considering using a mouse-blocking to reduce any potential high background signal.

Thank you for your email. As a positive control I can recommend to use sections from breast, placenta, kidney, liveror pancreas. Indeed, the prolactin receptor is expressed in those tissues. (http://www.uniprot.org/uniprot/P16471) There should however also be an expression in the salivary gland, as seen on the datasheet of ab2772. We do have in our catalogue tissue slides which can be used as positive controls. Dear abcam, A few months ago We order's primary antibody and one of them is Prolactin Receptor Antibody (U5) catalog ab 2772. Our order number xxx We have problem about Positive control, we use gland parotis tissue for Positive control and we used method IHC-P, but we can't found Positive Result. We have repeated this procedure three times, but We still did not give positive results. Could You kindly give us advise about our problem? Thank You very much. Looking forward hearing your reply.

We take product complaints very seriously, and investigate every product that we feel may not be performing correctly. Our products are covered by our guarantee. We will therefore look together through the protocol, and try to find a solution to the problem. If the vial would be faulty, we would replace it for you. In order to proceed, I am attaching our questionnaire so that we can gather further information regarding the samples tested and the protocol used. As shown on the Abreviews (feedback obtained from end users), the prolactin receptor antibody ab2772 has been successfully used in IHC-Fr on Mouse skin sections and in IHC-P on Human salivary gland sections fixed with paraformaldehyde : http://www.abcam.com/index.html?pageconfig=reviews&intAbID=2772 .

For IHC-P, the antibody was used at a dilution 1/100, for 12 hours at 4°C. No antigen retrieval method was applied.

Another positive control recommended for alternative prolactin receptor antibodies is breast carcinoma.
