Application notes
(see key)

Recommended dilutions
WB: 1/2000. Detects a band of approximately 65 kDa (predicted molecular weight: 31 kDa). A fusion protein was used in this blot, hence the difference in M.Wt as compared to native protein.

Not yet tested in other applications.
Optimal dilutions/concentrations should be determined by the end user.

Relevance

QPRT is a key enzyme in catabolism of quinolinate, an intermediate in the tryptophan-nicotinamide adenine dinucleotide pathway. Quinolinate acts as a most potent endogenous exitotoxin to neurons. Elevation of quinolinate levels in the brain has been linked to the pathogenesis of neurodegenerative disorders such as epilepsy, Alzheimer's disease and Huntington's disease.

Western blot - QPRT antibody (ab37515)

QPRT antibody (ab37515) at 1/2000 dilution + E. coli-derived fusion protein at 2.5 µg

Secondary
Goat anti-IgY-HRP at 1/1000 dilution

Predicted band size : 31 kDa
Observed band size : 65 kDa (why is the actual band size different from the predicted?)

Band size larger than expected as a fusion protein was used as positive control.
